List down the step-by-step procedure in AST:
- Prepare your pure inoculum using sterile distilled water/NSS. Add 3-5 colonies from your source plate and add it into the tube containing sterile distilled water/NSS. Match its turbidity with the McFarland standard.
- Dip a sterile swab into the pure inoculum. Press the swab into the sides of the tube to remove excess fluid.
- Inoculate the MHA using the lawn culture method. Using the swab, make a cross across your MHA plate. Then inoculate it from 3 directions to make a lawn. Pass the swab around the edge of the agar surface 3 times.
- After 5 minutes, place the antibiotic discs along the surface of the plate using sterile forceps. Make sure the discs are spaced evenly, with approximate distance of 15 mm from the edge of the plate. Gently press the discs on the agar to ensure contact.
- Within 30 minutes of preparation, incubate the plates overnight.
- Measure the zone of inhibition using a vernier caliper or ruler.
- Interpret the zone sizes using the chart.
