Laboratory 4 Flashcards
What are you doing this week?
(2)
Observing protein profiles made last week
Gel observation and size measurement of GFP
What did you use to de stain your gel?
Acetic acid/methanol/H2O
What is RF?
The distance travelled by the dye front relative to the migration of the various protein markers
How do you determine RF?
Distance travelled by the protein band / Distance travelled by the dye front
How do you measure RF?
Measure in centimetres or millimetres
Measure to the middle of the protein band
What do you graph?
(3)
Plot RF against the log of the molecular weight of the standards
Make standard curve
Derive molecular weight of the GFP by reading off the standard curve
What must you remember to do when reading off the graph?
Convert reading to molecular weight using the Anti-Log function on calculator
Explain in your own words how you work out each RF
(2)
Measure how far the dye front travels
Measure how far your first protein marker travels, divide this by how far the dye front travels
What do you find the log of?
The molecular weights of your standards -> these will be given to you
