What was the set up for this experiment?
A
A single colony from the agar + ampicillin plate was cultured overnight in 3ml LB broth in a shaking incubater at 37 degrees

Why is GFP not expressed in the agar + ampicillin plate?

A
These cultures do contain the plasmid as they are ampicillin resistant
But without arabinose the GFP is under negative regulation of the Ara-C protein (ara-C is the regulator)

Stock arabinose is 250mg/ml, we want 500 ug/ml

How did you carry out this experiment?
A
Spin 1 ml of culture. remove supernatant
Store pellet at -20 degrees
Add arabinose
Take 1ml of culture and spin as before
Repeat procedure for different time stamps T15, 30, 60, 90
Grade Green fluorescence of pellet on the UV box
