LAB 9 - Enrichment and Enumeration of Pollutant Degrading Bacteria Flashcards
1
Q
Enrichment culture
A
- Helps in isolating bacteria that are relatively rare in a sample
- Enrichment culture provides growth conditions that are favourable for the organism of interest but unfavourable for competing organisms
2
Q
Enrichment of thermophilic bacteria
A
Incubation at high temp (>55ºC)
3
Q
Enrichment of halophilic bacteria
A
Nutrient media supplemented with NaCl to encourage growth
4
Q
Enrichment based on nutrient conditions
A
Isolation of bacteria that can degrade organic chemical pollutants (e.g. oils and pesticides) by use of a growth medium where pollutant in question is included as essential growth constituent (e.g. sole source of carbon)
5
Q
Example of enumeration technique
A
- Most Probable Number
- Sample is serially diluted (usually 10-fold) in liquid culture medium, and must be diluted enough times so that the final (most diluted) tubes show no growth
6
Q
Result of complete microbial degradation (oxidation) of oil
A
Production of CO2, H2O, and microbial biomass that enters natual food webs
