Solution II containing sodium dodecylsulfate (SDS) and sodium hydroxide (NaOH)
SDS is a detergent that denatures proteins and disrupts the plasma membrane causing the cell to lyse(released into solution)
NaOH increase pH of the lysate and denatures the DNA
- Genomic DNA strands are able to move a greater distance
Solution III (acidic potassium acetate solution)
Causes some of the hydrogen bonds in the genomic DNA to re-form
- random pairing of complementary bases to create a tangled, insoluble mass of DNA
- acts as the salt in the banana DNA extraction
Ethanol in the supernatant solution
Removes water molecules from DNA
- the sodium ions and ethanol cause the plasmid DNA and RNA to come out of solution and precipitate
Solution T ( Tris buffer)
Any residual DNAese left in the preparation would be able to degrade the plasmid DNA

Approx 4000 genes in total
- arranged in a circle that is anchored to the bacterial plasm membrane
- kind of looks like squiggles

Float freely in cell’s cytoplasm
