Solution II containing sodium dodecylsulfate (SDS) and sodium hydroxide (NaOH)

A

SDS is a detergent that denatures proteins and disrupts the plasma membrane causing the cell to lyse(released into solution)
NaOH increase pH of the lysate and denatures the DNA
- Genomic DNA strands are able to move a greater distance

Solution III (acidic potassium acetate solution)

A

Causes some of the hydrogen bonds in the genomic DNA to re-form

  • random pairing of complementary bases to create a tangled, insoluble mass of DNA
  • acts as the salt in the banana DNA extraction
