Medium
Solution or solid that contains nutrients cells can use to grow and divide. It can be left as a liquid or can be mixed with a substance to solidify it.
Ex: calcium chloride solution
Colony
Small mass of genetically identical cells
Pure Culture
Isolated group of genetically identical cells
- use aseptic technique to avoid contamination
Contrast
Difference between light and dark areas on the slide
- can make untainted or translucent specimens stand out against the background
Magnification and Total magnification of the microscope
On the side of each lens
40/0.65= lens magnifies an image 40 times its actual size and has a numerical aperture of 0.65
Total Magnification=product of the magnification of the objective lens and the ocular lens
Ocular micrometer
Measuring scale in the ocular lens(eyepiece)
Parfocal lens
lens that stays in focus when magnification/focal length is changed. There is inevitably some amount of focus error, but small enough to be considered insignificant
Resolution
Capacity to distinguish two adjacent points as being distinct
Constant
Factors which are kept constant throughout the experiment
Experimental group
Same as control group with the exception of one condition or variable that is being tested
Will examine the effect of the independent variable on the dependent variable
Control group
Dependent variables in the absence of the independent variable
Solvent control group
Examines the solvent used in the experiment
- confirms the effect on the dependent variable you are measuring is due only to the independent variable and not the solvent dissolved in
Hypotonic
Has a lower solute concentration that the cell
- the cell will fill with water and swell becoming turgid
Hypertonic
Has a higher solute concentration that the cell
- water will leave the cell and plasma membrane will pull away from the cel wall called plasmolysis
Isotonic
Has solute concentration the same as the cell
- no net movement of water which causes the cell to become flaccid b/c the reduced pressure on the cell wall
