How do you calculate total magnification?
Ocular lens power X objective lens power = total magnification
How should you carry a microscope?
With two hands - by the base and arm, don’t tilt
What should you use to clean parts of a microscope?
Kim wipes
What magnification should you start with?
4X
When you leave the classroom, how should you leave the microscopes?
On the lowest objective, power off, and cord wrapped neatly
What is a cross section?
made perpendicular to the longest axis
What is the longitudinal section?
made parallel to the longest axis (along the longest axis)
How do you calibrate the field of view?
Focus on your calibration slide at 4X, match the width of the FOV with the width of the lines, measure at 6 different points and record estimates, and repeat for the 1X & 40X - average all estimates
What should you do if your scope lens becomes dirty?
Ask the TA to come clean it
Light microscope (LM)
uses light and magnification to see something small
Transmission electron microscope (TEM)
a microscope technique that reads thin specimens of about 0.2 nm - uses electrons
Scanning electron microscope (SEM)
an electron microscope that scans the surface of a solid with electrons - 3D image up to 50,000X
