How were HSC identified?
A
TP bm of healthy mice into irradiated mice = colonies on spleen of mice. These colonies could be tp to another irradiated mouse to form new colonies. This suggested the presence of colony-forming units.

What are the requirements for master regulatory TFs?
A
- Required for development into a specific lineage
- Can change the fate of cells they are introduced into
- Antagonise the opposite lineage programs

Give 2 examples of master reg of HSC and their roles
A
GATA1 and PU.1
GATA1 - req for eryth dev
PU.1 - req for myeloid dev
Both TF inhibits the other therefor no net expression of a particular lineage set of genes = uncomm/undiff cell line
