hyperlipidaemia Flashcards
What drug class is Atorvastatin?
Statin
What are the common indications of Atorvastatin?
Hyperlipidemia
What is the mechanism of action of Atorvastatin?
HMG co-A reductase. Inhibits the production of cholesterol. Enhances LDL receptors in hepatocytes.
What are some adverse drug reactions of Atorvastatin?
Rhabdomyolysis
What are some important drug-drug interactions or Atorvastatin?
Amlodipine, Amiodarone, Diltiazem (calcium channel blocker) and macrolides
What image can you use to remember Atorvastatin?
ATOR
Fit so lowers cholesterol
Adverse reaction- rhabdomyolysis (sword fights, injuries, muscle breakdown, blood)
when would you want to avoid taking a statin or a cholesterol lowering medication
when you are pregnant as baby needs cholesterol
what are some examples of drugs in the drug class fibrates
fenofibrate
ciprofibrate
what are the action of fibrates
they activates
PPARa to increase lipoprotein lipase production
so increased clearance of TAG and fatty acid uptake in the liver
what is an example of a drug that fits in the antilipolytic class
Niacin
It isn’t first line
what is the moa of niacin
reduces VDL and LDL
it increases HDL
and reduces triglyceride synthesis
what drug class is ezetimibe
NPC1L1 TRANSPORT INHIBITOR
how does ezetimibe work
acts at the brush borders and inhibits NPC1L1 Transporters
so they reduce the absorption of cholesterol in the small intestine
they also increase hepatic LDL receptors so more gets taken up
what is the risk of giving ezetimibe or fibrates with a statin
it causes rhabdomyolysis
what is the mechanism of action of Alirocumab or evolocumab
they are PCSK9 inhibitors
