Hematocytometer Flashcards
Hematocytometer/cell counting chamber
Counting chamber
Used for manual counting blood cells
Hematocytometer invented by
Louis Charles malassez
Hematocytometer consists of
Thick glass microscope slide
Rectangular identation create precision volume chamber
Principle of hematocytometer
No of cell counter=sum of cells counted across square in one chamber
Different types of counting chamber
1,Thoma cell counting
2,malassez
3,Improved new vauer
4,burger
5,fuch rasanthal
Thoma cellcounting
Contain a central square with 1 mm with 10x objective
*for ruling leukocytes and hemoglobin
Chamber Used to count nematodes from cerebrospinal fluid
Malassez
Principle of malassez
No of particles per unit volume
For particles within the particles (leucocytes, erythrocytes)
Improved newbauer
3mm ×3mm area
9 squares with 1 mm each
4 corner square to Determine WBC count which 8s subdivided into16
*to determine RBC ,4 corner squares and central squares(1/5) of 25 subdivided portion of the central square is used
Covers slip for counting chamber
Can count under low power and high power but not for oil immersion
3 thickness and 2 sizes
Thickness-0.3,0.4(common),0.5
Size-16×22 (single ruled) and 22×23(double ruled)
Changing counting chamber
Raised shoulder of chamber and coverslip should fit properly -to ensuredepth of the chamber is uniform
*the diluted blood should does not overflow to the H shaped trough, so are must be needed
*drop added to edge of cover blast and drawn by capillary action
*for counting platelets, simple moist chamber by placing a cotton/wet pad on petridish can be used
*before counting, allow cells to settle in the moist chamber
