Streak Plate Method of Isolation
the goal is to achieve isolated colonies or colony forming units
-ASEPTIC TECHNIQUE required
Colony
a visible mass of cells on a petri dish arising from a single cell
-will have genetically identical cells since they originate from a single cell
Dilution gradient
taking from high concentration to low concentration; carries fewer and fewer cells into the subsequent areas of streaking
Colony Characteristics
shape (form), margin, elevation, texture, surface, color
Shape
circular, irregular, punctiform (tiny)
Margin
entire (smooth), undulate (wavy), lobate (lobed), filamentous or rhizoid
Elevation
flat, raised, convex, pulvinate (very convex) and umbonate (raised center)
Texture
moist, mucoid/slimy, dry
Surface
rough, smooth, glistening, wrinkled
Color
may be combined with optical properties such as opaque, translucent, shiny or dull
