- founding father in microbiology
- introduced the concept of pure cultures and growing microorganisms in culture media
Macronutrients
nutrients needed in large amounts such as carbon and nitrogen
Micronutrients
nutrients needed in very small amounts
-ex: trace elements (cofactors; Iron, Cobalt, Copper, etc) and growth factors (vitamins, amino acids, other organic molecules)
Agar
solidifying agent in microbiological media since very few non-marine bacteria hydrolyze it
-extracted from red algae
Tryptic Soy Agar (TSA)
used to make agar plates, deeps, and slants
Tryptic Soy Broth (TSB)
used for broth cultures
Defined medium
will contain measured amounts of salts and nutrients of known composition
Complex medium
the exact composition in not known
All-purpose medium
provides the nutritional requirements of a wide range of bacteria
Selective media
media with added compounds that help promote the growth of a selected few types of bacteria
-ex: NaCl, antibiotics, dyes, specific inhibitors
Differential media
media with certain compounds that allow them to differentiate between closely related organisms
Different forms of media
- liquid cultures
- semi-solid media (0.4% agar)
- solid agar (1.5% agar)
Autoclave
machine similar to a pressure cooker that uses heat and steam to achieve sterilization
-sterilization is achieved when all living cells including bacterial endospores are dead
Filtration
sterilizes heat labile (sensitive) materials like antibiotic solutions and urea broth
Ethylene oxide
sterilizes heat labile (sensitive) materials
