False Negative Reactions
Can occur when antigen-antibody reactions have occurred but WASHING IS INADEQUATE & free antibody remains when the AHG is added (inadequate cell washing, delay in adding AHG after washing, presence of small fibrin clots among cells, inactive/forgotten reagent, shaking too hard
False Positive Reactions
Using improper sample (not EDTA), spontaneous agglutination (cells heavily coated with IgM), non-specific agglutination (sticky cells)
Function of check cells
to make sure the AHG was added after wash & make sure reagent is working
Elution
procedure that removes antibody bound to the RBC, this antibody goes into the eluate, which can be used for serologic testing
Adsorption
process of adding specific antigen so that antibody can attach to it & be removed from a serum, the antigen adsorbs the antibody
Absorption
process of removal of antibody from a serum
IAT
RBC antigen typing, antibody detection and ID, Weakened D test, crossmatch… detect In vitro sensitization of red cells
DAT
Detects In vivo coating of patient cells (IgG antibodies, complement, or both)
Purpose of pre-transfusion testing
Ensures ABO compatability between the donor and the recipient and should detect most clinical significant red cell alloantibodies that will react with donors red cells
