Exam 1 Lecture 6 Flashcards
Pharmaco dynamics
-What does a drug do at its site of action
Ligand types
-Agonist
-Antagonist
Agonist
Activates receptor
Antagonist
-Inactivates receptor
-Competitive: Binds to regular binding site, competes with agonist
-Noncompetitive: Binds to different binding site
Partial agonist
-Binds to a receptor and activates it, but not to its full effect
Dose response curve
-Measures how much dose to get 50% effectiveness: ED50
Dose response curve antagonist
-Stays at 0 because it inactivates receptor
Dose response curve with agonist and competitive antagonist
-ED50 shift to the right
-Need more dose to reach ED50 (because fewer receptors to bind to)
-Emax is unchanged
Dose response curve with agonist and non competitive antagonist
-ED50 stays
-Emax reduces (because some receptors don’t work)
Dose response curve with agonist and partial agonist
-ED50 stays
-Emax reduces (because partial agonist have weaker response)
TD50
LD50
-50% individuals receive toxicity
-50% individuals die
-drug can be 0% lethal at 100% response (good)
-Can be 50% lethal at 70% response (not good)
Therapeutic index
-Diff between LD50 and ED50
-Large = safe
-Small = unsafe
-Helps with range its safe to use drug
ED50
How much agonist is needed to reach 50% of full effect
Withdrawl
-Physical and psychological symptoms when someone stops using a drug
Sensitization
-Body becomes more sensitive to the drug
-More receptors available
-Body will do this in response to withdrawl
Tolerance
-Body becomes less responsive to drug/ weakens it
-Down regulates receptors available
-To keep homeostasis and protection
