What are the characteristics of Amylin receptor agonists?
● MOA : Acts on amylin receptors in the hindbrain or by a vagally mediated mechanism to:
● Slow gastric emptying
● Suppress postprandial glucagon concentration
● Increases Satiety (i.e., decreases appetite)
Which are the Amylin receptor agonist drugs?
Pramlintide ( Symlin )
What are the characteristics of Pramlintide ( Symlin )?
● Type1/Type2diabetics who use insulin at mealtime and have failed to achieve diabetic control
● Injected S.C. before meals (reduces postprandial glycemia)
● Weightloss
● S.E. =
● Nausea (28-48%)→titrate dose to avoid
● Vomiting, anorexia, headache
● Hypoglycemia
● Caution in patients with diabetic gastroparesis
● Drug interactions because it slows gastric emptying →Don’t take with other drugs within 1 hour before or 2 hours after injection. Don’t coadminister anticholinergics, (that may slow gastic motility ) acarbose or miglitol
● Half-life = 48 minutes
● No hypoglycemia if used alone, but increases risk of severe hypoglycemia if used with insulin
