What is a random sequential mechanism?
Substrate and enzymes bind, forming ternary complex, but the order of binding/ release doesn’t matter.
What is an ordered sequential mechanism?
When there is a specific order to the substrates and enzyme binding and the product produced.
What is AZT?
A drug that inhibits replication of HIV.
It is a nucleoside analogue.
What is a transition state analogue?
An inhibitor that looks like the transition state.
What is a catalytic antibody?
Antibodies that bind a transition state, causing catabolism.
What is a concerted model?
(Alloteric) enzyme complex where all of the active sites are either open or closed.
What is a sequential model?
An allosteric enzyme complex where a change in one of the active sites changes the others.
What is proteolytic cleavage?
Inactive molecule that is cleaved, making it active.
What comes with a competitive inhibitor?
An increase in Km (its harder to bind)
No change in substrate (Can be overcome with an increase in substrate concentration)
What comes with a non-competitive inhibitor?
The km is unchanged (substrate can still bind) Max decreased (substrate rise cannot overcome this)
What is Km a measure of?
The affinity of the enzyme and substrate for one another.
What are assays?
Lab methods for studying enzymatic activity.
