alpha-1 blockers side effects
GI: xerostomia and nausea
eye: “floppy iris syndrome”
CNS - dizzy, somnolance, asthenia, insomnia
alpha-1a specific blockers
tamsulosin and silodosin
tamsulosin and silodosin adverse effects
abnormal ejaculation, dopamine and other neurotransmitter blockade
tadalafil moa
inhibition of PDE-5 decreasing the breakdown of cGMP resulting in prolonged high levels of cGMP, resulting in sm. muscle relaxation
tadalafil adverse effects
rare - optic neuropathy, retinal artery occlusion, hearing loss;
contraindicated with concurrent organic nitrates
finasteride and dutasteride moa
inhibition of 5alpha reductase, blocking the conversion of T to DHT, preventing the DHT driven proliferation of the prostate
finasteride (type II 5-alpha reductase)
dutasteride (type I and II 5-alpha reductase)
5-alpha reductase inhibitors adverse effects
erectile and ejaculation dysfunction, decreased libido, gynecomastia, decreased PSA, teratogenic (but not carried in semen)
least useful alpha-1 blocker
prazosin due to its short duration of action
