Main difference between first-generation and second-generation anti-histamines
A
First-gen have more side-effects such as anticholinergic, anti- alpha adrenergic (postural dizziness, falls) and anti-serotonergic (appetite stimulation, weight gain)
First gen: chlorpheniramine, diphenhydramine, hydroxyzine, promethazine.
Second-gen do not have the same degree of antimuscarinic, antiserotonergic, and antiadrenergic properties and their side effects are minimal.
Second gen: loratadine, cetrizine
