What is distribution?
The dispersion of a drug throughout your body
from: vascular space to: extravascular space( fat, muscle, nervous system)
How does the concentration equation change when looking at distribution?
C(ideal)= total mass absorbed/ Volume (which the drug is distributed)
-the volume includes the plasma and extravascular space
What does IV bolus: Concentration vs time graph with distribution (no met or excretion) look like?
What is the Concentration of the drug in plasma with low Vd?
if drug stays in plasma then it will have a high concentration.
ex. plasma protein bound, H2O soluble, large MW
What is the Concentration of the drug in plasma with high Vd?
if drug gets widely distributed then the concentration will be decreased
ex. lipophilic, binds to extravascular tissues, small MW
What are some determinants of Vd?
- properties of the drug: can the drug get through the vascular ensothelium and into extravascular space?
- size of extravascular space: estimated from weight of people. obese will have larger vascular & extravas space.
how do you calculate the Apparent volume of distribution?
Vd (L/kg) * weight (kg)
Does Warfin have a high or low Vd?
high degree of plasma protein binging-stays mostly in blood. low Vd
Does Gentamicin have a high or low Vd?
low Vd. large water soluble molecule- stays in extracellular water
Does EtoH have a high or low Vd?
small water soluble molecule- it goes where water goes
Does Chloroquine have a high or low Vd?
high Vd. highly lipophilic. very little stary in vascular compartments–>gets into fat and most tissues
