What sort of drugs cross the BBB easily?
Light weight (molecule weight) lipophilic drugs can cross the BBB radpily.
Large drugs are excluded
Give examples of drugs that can cross the BBB easily
NSAIDS and paracetamol (And any illegal ones) can cross the BBB easily and are rapid acting
what drugs cant get into the brain?
Proteins
(large drugs)
Monoclonal antibody therapies
Unless theres a specific transport mechanism these drugs are blocked.
What receptor does caffeine act on in the brain?
Adenosine R antagonist
What receptor does alcohol act on?
NMDA and GABA R
What receptor does cocaine act on?
Blocks DA transporter
Whats the difference between CB1 and CB2?
The endocannabonoid receptors:
CB1= brain specific, highly expressed
CB2 = leukocyte specific
What does CB2 activation lead to?
CB2 activation protects against cerebral ischemia by inhibiting neutrophil activation
Why target CB2?
It is only found on immune cells and by preventing them from crossing the BBB prevents the BBB destruction and harm caused by inflammation in the brian.
What disease would CB2 activation be useful?
MS which is an autoimmune disease therefore inhibiting what causes it (neutrophils is handy)
What sort of receptors are CB1 and CB2?
GCPR
What targets CB1?
THC (plant dereived)
Endogenous ligands = 2AG and Anandamide
What sort of receptors is CB1R expressed in?
GABA
Glutamate

To prevent neutrophil migration into the brain what could be targeted in MS?
The CB2 expression on the nueotrophils and the molecules they interact with on the vasculature of the brain.
