Culturing terminology and streak plating quiz Flashcards
what is a culture?
the growth of bacteria in a lab setting
what is inoculum?
placing bacteria in a growth medium
what is a growth medium (media)?
supplies nutrients and energy requirements for microbial growth
what are examples of the types of nutrients growth medium would contain?
carbon source - glucose
nitrogen
vitamins
water
what are the two forms media come in?
solid
liquid
what is another name for solid media?
agar
what is another name for liquid media?
broth
what are the three forms agar can be displayed?
angled in a test tube (slant)
flat top in a test tube (stab)
in a petri dish (plate)
what is the standard solid media for growth?
Tryptic soy agar (TSA)
what does tryptic soy agar contain?
tryptones -(amino acids)
soy contains: glucose, nitrogen and salts
How can liquid media be displayed?
can be clear -> indicating no growth
can be cloudy (turbid) -> indicates there is growth
what is the standard liquid media for growth?
tryptic soy broth (TSB)
what is the goal of streak plating?
to isolate one type of microbe
what were the bacteria we used in our streak?
Staphylococcus aureus
Escherichia coli
what was the first step of streak plating?
take one loopful of bacteria and spread over 1/4 of plate
what do we do between each main step of streak plating? Why?
flame the loop
to reduce the number of microbes as we progress each step
what was the second main step of streak plating?
after flaming loop, start in zone 1 and spread bacteria over another 1/4 of plate
what was the third main step of streak plating?
after flaming loop, start in zone 2 and spread bacteria over another 1/4 of plate
what was the fourth main step of streak plating?
after flaming loop, start in zone 3 and spread bacteria last 1/4 of plate, leaving a tail in between (but not touching) all the other zones
