What is plasmolysis?
A
- Protoplasm of plant cell begins to shrink away from cell wall.

What is incipient plasmolysis?
A
- Where water potential of cytoplasm is same as solution.
- Point at which plasmolysis just begins to occur.
- 50% of cells in tissue are plasmolysed.

Why are plant cells left in sucrose solution for 20 mins before observation?
A
- To allow time for osmosis to reach equilibrium.

Outline the procedure of this practical?
A
- Create different conc. sucrose solutions + place in watch glasses.
- Place thin layer of onion tissue in each solution ensuring submersion.
- Leave for 20 mins.
- Remove using forceps.
- Mount in drop of corresponding solution on slide + add cover slip.
- Observe cells + count how many are plasmolysed.
