What is incipient plasmolysis?

A
  • Where water potential of cytoplasm is same as solution.
  • Point at which plasmolysis just begins to occur.
  • 50% of cells in tissue are plasmolysed.

Why are plant cells left in sucrose solution for 20 mins before observation?

A
  • To allow time for osmosis to reach equilibrium.

Outline the procedure of this practical?

A
  • Create different conc. sucrose solutions + place in watch glasses.
  • Place thin layer of onion tissue in each solution ensuring submersion.
  • Leave for 20 mins.
  • Remove using forceps.
  • Mount in drop of corresponding solution on slide + add cover slip.
  • Observe cells + count how many are plasmolysed.
