Area under the curve.
Which two pathologies would alter clearance?
Pathology altering perfusion of the organ involved (e.g. HF = less blood to organ)
Pathology altering the organ function (damage to organ)
Fub stands for?
Fraction of unbound drug.
I.e. fraction of the drug available to be metabolised
Explain CL-int
The intrinsic ability of the enzymes (e.g. hepatic enzymes) to metabolism the drug.
For drugs with low hepatic clearance, what is the rate limiting step of the clearance? CL-int or Fub?
