Chapter 3 Flashcards
Isolated colonies
Are separated from each other
Colony morphology
Each microorganisms exhibits certain phenotype: SHAPE (round irregular pinpoint) MARGIN (undulate) ELEVATION (flat) TEXTURE (moist) PIGMENT PRODUCTION (shiny dull)
Environmental factors include?
Oxygen, temperature, osmotic pressure, pH
Oxygen types
OBLIGATE (strict) AEROBIC, requires presence of oxygen for growth, OBLIGATE (strict) ANAEROBIC, can’t grow in presence of oxygen, FACULTATIVE ANAEROBIC, grow in the presence or absence of oxygen but prefer oxygen, MICROAEROPHILLIC, requires oxygen, but at lower atmospheric concentrations (2-10%) greater than 10 toxic
Anaerobic jar
This device will provide anaerobic environment free of O2 include: GAS PACK, PALLADIUM PACKETS, AND INDICATOR STRIPS
Candle jar
Great for microaerophillic organisms, candle flame will consume oxygen in jar to about 2-5% produce elevated CO2 levels
Fluid Thioglycollate Medium (FTM broth)
Determines growth characteristic of organism based on oxygen requirements. The reducing agent is THIOGLYCOLIC ACID creates anaerobic environment as tube gets deeper, METHYLENE BLUE/RESAZURIN redox indication turns pink in presence oxygen and colorless when oxygen absent
