Q

Describe the magnetic separation technique used on teh centaur.

A

during incubation, coated PMP in the cuvette binds to the target Ag or Ab. When the cuvette is exposed to a magnetic field, the PMP bound to Ag or Ab are drawn toward the magnets. While the magnets hold the PMP in place, sample and reagent not bound to the coated PMP are washed away.

7
Q

Describe the sandwich assay reaction.

A

Uses AE-labeled Ab in the lite reagent. Acid-base are added to initiate the chemiluminescent reaction. The emission of light is measured in relative light units (RLUs). Once the light produced from the oxidation of Acridinum Ester is quantitated and the system calculates the concentration of antigen.

Q

Lite reagent, containing AE-labeled Ab, and solid phase, containing Ag-labeled PMP are added to the sample. the cuvette is incubated at 37C. Ag bound to PMP competes with analyte-specific Ag in the sample for limited binding sites on AE-labeled Ab.
