Centaur Flashcards
Principle of direct chemiluminescence on the centaur
Direct chemi. reaction directly measures the light energy without the use of added steps or amplifying molecules.
It uses _____as the chemiluminescent label
acridinium ester
Describe the magnetic separation technique used on teh centaur.
during incubation, coated PMP in the cuvette binds to the target Ag or Ab. When the cuvette is exposed to a magnetic field, the PMP bound to Ag or Ab are drawn toward the magnets. While the magnets hold the PMP in place, sample and reagent not bound to the coated PMP are washed away.
The proportionality between chemiluminescence and analyte concentration: Sandwich
Direct proportionality between the analyte Ag concentration in the sample and light emission in relative light units.
The proportionality between chemiluminescence and analyte concentration: Competitive (with AE-labeled antigen and AE_labeled antibody)
inverse relationship
The proportionality between chemiluminescence and analyte concentration: Antibody capture
Direct relationship
Describe the sandwich assay reaction.
Uses AE-labeled Ab in the lite reagent. Acid-base are added to initiate the chemiluminescent reaction. The emission of light is measured in relative light units (RLUs). Once the light produced from the oxidation of Acridinum Ester is quantitated and the system calculates the concentration of antigen.
Lite reagent, containing AE-labeled Ab, and solid phase, containing Ag-labeled PMP are added to the sample. the cuvette is incubated at 37C. Ag bound to PMP competes with analyte-specific Ag in the sample for limited binding sites on AE-labeled Ab.
Competitive
If sample has ___concnetration of analyte-specific Ag, most binding sites on the Ab are bound to Ag from the sample, and ____sites are bound to PMP labeled Ag. Which results in a ____ reading of ___from the oxidation of AE.
Increased, few, lower, RLUs.
Used when the substance being measured in the sample is an Ab. This assay can detect specific IgG Ab or IgM Ab in patient sample.
Antibody capture.
What is the purpose of 1st incubation and wash in antibody capture?
to remove excess interfering substances in the sample.
What is the purpose of second incubation and wash in antibody capture?
measure the Ab in the sample.
If more sample IgM Ab is present, then ____ is present, therefore the light emission is ___
more AE, higher
