Advantage
1. Electron wavelength longer
2. TEM has higher resolution
3. Small objects can be seen
Limitations
1. Cannot look at living cells
2. Must be in vacume
Scientists isolated mitochondria from liver cells. They broken open cells in a ice cold buffered isotonic solution.
Why ?
- Prevent osmosis so organelle doesn’t burst
- Reduce enzyme activity so organelles aren’t damaged
- Maintain constant PH so protein don’t denature
Describe and explain how cell fractionation and centrifugation can be used to isolate mitochondria from animal cells
- Cell homogenisation to break open cells and release organelles
- Filter to remove whole cells
- Use isotonic solution to prevent osmotic damage to organelles
- Keep cold to prevent enzyme damage
- Use buffer to maintain ph
- Use differential centrifuge to separate nuclei
- Respin at higher speed to get mitochondria in pellet
