antivirals Flashcards
Acyclovir
Nucleoside analogue.
Acyclovir resistance
mutation of the thymidine kinase gene. Cross resistance w/ other antivirals with similar mechanism
Acyclovir adverse effects
nausea, headache, diarrhea. Reversible crystalline nephrotoxicity. Maintain hydration
Valacyclovir
Prodrug of acyclovir. 3-5x greater oral bioavailability than acyclovir
Foscarnet
Does not require phosphorylation step as in acyclovir.
Ganciclovir
requires viral kinase to be activated
Resistance go ganciclovir
Mutation of the UL97 gene is most common
What is prodrug of ganciclovir
Valganciclovir
What is most common complication of influenza
pneumonia, either primary viral or secondary bacterial pneumonia
mech of action of oseltamivir
Binds active site of neuraminidase to inhibit its function. Inhibits spread of progeny virions thru the respiratory tract
What does zanamivir treat and how is it administered
Influenza antiviral administered via inhalation
What does peramivir treat and how is it administered
influenza, via IV
Amantadine and Rimantadine are effective against what strain of influenza
Influenza A M2 prtn
Which drugs act on HVC NS3/4A
The protease inhibitors paritaprevir and simprevir
Mutation leading to resistance of paritaprevir
Specific NS3 gene substitutions can render the virus resistant
