Culturing microroagnisms
Inoculation: Introduction of a new sample into a container of growth media
Borth culture
Is a liquid growth media allowing bacteria to grow. They are inoculated with bacteria then the are incubated to allow the bacteria to multiply. They precipateta nd turn cloudy as the popualiton increases
Why are agar plates streaked
This si to spread out the bacteria so that oindividual colonies can grow
Cell counts
Stain broth smaple with tryptan blue in order to stain dead cells blue
Calibrate the haemocytometer with a volume of 0.1mm3. Count the number of cells within each set of squares and then calculte teh eman
NUmber fo bacterial cells= Number counted x 10^4
Dilution plating
Grow colonies from single microorganisms
Perform serial dilutions with distilled water in order ot see sinlge colonies
Prepare lwn and count number of colonies
NUmber of cells = NUmber of colonies x dilution factor
How is staphyllococcus ssp caused?
Endotoxins are released as the bacteria metabolises and reproduces. The endotoxinsspread around the body in the blood and tissue fluids
How does salmonella ssp casue disease
Lipopolysaccaride endotoxins on outer clell mmebrane of gram negative bacteria. THe endotoxins are released when the bacterium dies
Mycobacterium tuberculosis
Mycobacterium tuberculosis infects a phagocyte. The phagocyte is enclosed in a tubercle as a result of the inflammatory reponse in the lungs. The bacteria remains dormant in the tubercle and is not affected by the immune reponse due tothe tubercle contianing a thick waxy coat. When the immune system is weakned, the bacteria beocmes active again and detsorys the lung tissue thus causign tuberculosis or coughing problems weight loss and fever.
