ames test quiz Flashcards
What is the ames test
whether a chemical can mutate DNA
Mutagen
a chemical substance or physical agent that can induce changes in the base sequence of DNA
Carcinogen
mutation that can cause cancer
Types of base mutations
silent, nonsense, missense
silent
change in a base that will still code for the same amino acid
nonsense
codon will code for the sequence to stop
missense
code for another amino acid
frameshift mutation
nucleotides are inserted or deleted, shifting the sequence
what type of mutation did the M starin have
a missense mutation in the histidine gene
mutation that f strain has
deletion frameshift mutation
for a mutation to occur on the media without hist, the bacteria needs to have
back mutation
what is a back mutation
mutated genes that will revert back to their wild type if a second mutation occurs.
explain the experiment
m strain with missense mutation and f stain with frameshift mutation. both mutations inhibit the bacteria to grow. the strains lack the sequence to reproduce. these strains are placed mm plates with either NaAz(m strain) or 4-nitro ( f strain). The plates do not contain histidine. If colonies appear, these two chemicals cause a back mutation. The bacteria can now reproduce and colonies would appear. That means it tested positive, and the chemical can now be further tested to see if it’s a possible carcinogen.
sodium azide (NaAz)
is known to cause base mutations
4nitro
known t cause frameshift mutations
