analgesic and opioid receptor agonist
What are the uses of Alfentanil?
- rapid onset makes it ideal for analgesia when noxious stimulation is acute, but transient, as associated with laryngoscopy, tracheal intubation, and performance of a retrobulbar block
- analgesic adjunct in the maintenance of anesthesia with barbiturates or N2O or as primary anesthetic agent for the unduction of anesthesia
What is the MOA of Alfentanil?
mu-opioid receptor agonist
mu, kappa, delta
-drug binds to stereospecific receptors at many sites within the CNS, increases the pain threshold, alters pain perception and inhibits ascending pain pathways
What is the onset of action of Alfentanil?
RAPID- effect site equilibration time is 1.4 minutes
What is the DOA of Alfentanil?
30 - 60 minutes
How is Alfentanil metabolized?
N-dealkylation and O-demethylation in the liver to form inactive metabolites
What is the major metabolite? is it active?
Noralfentanil, it is NOT active
What is the elimination 1/2 life of Alfentanil?
83 - 97 minutes (1.5 hrs)
What is the Vd of Alfentanil?
4 - 6 times smaller than Fentanyl. This is due to lesser lipid solubility and higher protein binding
