Week 3 Flashcards
1
Q
What is an Endospore Stain?
A
a Differential stain used to visualize bacterial endospores
2
Q
What is an Endospore?
A
A thick-walled structure that protects the DNA of a bacteria in extreme unfavorable conditions
3
Q
how do you do the endospore stain?
A
- Prepare a heat-fixed slide with bacteria on it over a boiling water bath.
- Add a few drops of Malachite green to the smear then cover it with bibulous paper
- Keep on the water bath for 10-15 minutes and keep adding drops of malachite green so it doesn’t dry
- After removing the paper and rinse the slide with water
- Counter stain now with Safranin for 1 minute
- Rinse, Blot dry, and examine, start at 4x all the way to 100x oil immersion lense
4
Q
How long does it take an Endospore to form?
A
6-8 Hours
5
Q
What are spores resistant to?
A
Heat, Desiccation, Chemicals and radiation
6
Q
When spores are exposed to favorable conditions, they can germinate into a vegetative cell after how long?
A
90 minutes
7
Q
The normally-growing cell that forms the endospore is called a ?
A
vegetative cell
