What does enzyme treatment do?

A

Enzymes cleaves MNS enzymes and Duffy (Fya/b) antigens from cell surface so they cannot react in enzyme-treated cells.
The other antigens reactivity is enhanced as a result.

Identifying unexpected antibody set-up

A

Need 2x AHG gel (green) and NaCl gel (red).
Label with cell 1-11 + positive control.
1. Add 50uL of screening cell into each microtubule.
2. In positive control add 50uL of a RhD+ screening cell.
3. Add 25uL of test plasma into all microtubules (except control)
4. In +ve control add 25uL of weak Anti-D reagent.
5. Incubate at 37C for 15-20mins
6. Centrifuge for 10mins.
