A

How it works

A

Gram negative bacteria of decolourise by alcohol losing their primary stain purple

Gram-positive bacteria are not decolourise by alcohol and above remain as purple of the day colourisation start a counterstain is used to import the pink colour of the decolourise gram negative organisms

Method

A

Place slide with heat fix smear on staining tray

Flood smear with crystal violet one minute
Tilt slide gently friends with tapwater

Gently flood with iodine one minute
Tell slide gently rinse

Decolourise using acetone or alcohol Tilt slide slightly while doing so
Rinse immediately

Gently flowed with safranin to counterstain one minute
Tell slide and rinse

Blots slide dry

View the smear using a light microscope

Cheek sampling

A

Swab cheek with cotton bud

Wipe onto microscope slide

cover it with methodine blue

Put coverslip onto of cells

Check it in microscope starting with smallest magnification

Onion skin sampling

A

Peel thin layer of skin

Place onto microscope slide

Cover it was iodine

Put coverslip on top of cell

Check in microscope start with smallest magnification
