Amphetamines
psychostimulant. interfer with ability of synaptic vesicles to store monoamine. displace 5HT, DA, NE from storage vessicle increasing NE in prefrontal cortex. and inhibit monoamine oxidase.
ADV. drug of abuse
Atomoxetine
mood stabilizer. psychostimulant. selective NE reuptake inhibitor (increased NE in prefrontal cortex)
TX. ADHD
ADV cardiotoxicity. withdrawl could lead to suicidal (Taper), rare liver tox, less insomnia
Cocaine
psychostimulant. block reuptake (5HT, NE, DA), NA chanel blocker
ADV. drug of abuse. Tox vasoconstriction, mydriasis,
Withdrawl. depression
Dextroamphetamine
psychostimulant. interfer with ability of synaptic vesicles to store monoamine. displace 5HT, DA, NE from storage vessicle increasing NE in prefrontal cortex.
DOA: D isomer of amphetamine (similar to cocaine, noncathoaminergic)
Lithium carbonate
bipolar drug of choice. prevents recycling of inositol (decreasing PIP2 (Gq) and decreasing cAMP (Gs.i) )
DOA: narrow therapeutic index. similar to sodium (depolorize cells causing seizures)
TX. bipolar DOC. usually with antidepressant/antipsychotic in mania
ADV. seizures, endocrine (hypothyroidism TSH-Gs, peripheral deiodinase inhibitor thereby increasing depression), nephrogenic diabetes insipidus (nephro ADH-Gs) do not use thiazide which would cause increase lithium (sodium ike) therefor use amiloride or triampterien. teratogen epsteins anomoly.
Methylphenidate
Mood stabilizer. amphetamine-like (ritalin/concerta). causes downregulation of neurotransmitter.
TX. ADHD
ADV: agitation, restlessness, insomnia, cardiovacular toxicity (high NE). withdrawl could lead to suicidal (Taper)
Modafinil
psychostimulant. MoA unclear causes euphoria without abuse.
Tx. narcolepsy
