Isoniazid

A

Isonicotinylhydrazine (INH)

most potent anti-TB in drug regime

Bacteriacidal dividing (static in stationary phase). pro-drug activated by catalase-peroxidase (KatG)

covanlently binds and inhibits 2 enzymes in mycolic acid synthesis, a long chain beta hydroxylaated fatty acid.

  • enoyl acyl carrier protein reductase (InhA)
  • beta ketoacyl ACP synthase (KasA)

Oral. (do not take antacids or too much carbs at same time though). Met liver acetylation (person dependant)

ADV. peripheral neuritis (vit B6), hepatitis, convulsions, mental abnormalitis, HSR rash/fever, potentiation of drugs metabolized by p450.
