after centrifuging a sample the buffy coat is the thin layer between RBCs and plasma containing proteins

- methylene blue: stains acids blue (base stain that attracts acids)
- eosin: stains bases red (acidic stain that attracts bases)
- methanol: fixative
- phosphate buffer: maintains pH
