What is the point of this practical?
To investigate cells and see their size, what they look like and how they work
How do you work out Magnification
Image size divided by actual object size
What do you use to make the cell visible?
A coloured stain
What do you need to do with the specimen?
Make it as flat and thin as possible
How do you focus on the specimen?
Start on the smallest lens, focus then move up a lens.
How do you measure the size?
Use a ruler or eyepiece scale
1mm =?
1000um (micrometre)
1um =?
1000nm (newton metre)
Why can you see the nucleus and cell wall but not mitochondria?
They are far too small and not stained
How can you identify smaller parts of cells?
Use an electron microscope which has better resolution and magnification
Method:
- place tissue sample on the microscope slide
- add few drops of suitable stain
- Lower a coverslip onto the tissue
- place slide on the microscope slide and focus using low power
- change to high power and focus
- Draw types of cells that are observed
- Add scale line to diagram
