Required Practical - Investigating amylase enzyme Flashcards
1
Q
What is the point of the practical?
A
To find out what happens to the rate of enzyme activity when the PH changes
2
Q
Method:
A
- Put a test tube with a starch solution and a test tube with amylase into a water bath at 37 degrees.
- After 5 minutes add amylase to starch.
- Every 30 seconds take a drop from the mixture and test it for starch with iodine solution.
- Record how long it takes to become completely digested.
- Repeat the experiment with different PH values using different buffer solutions.
3
Q
What are the controlled variables?
A
temperature, concentration and volume of starch and amylase.
4
Q
What is the Independent variable?
A
PH value
5
Q
What is the dependent variable?
A
The time taken for the starch to be digested
6
Q
Why do you need a water bath?
A
To maintain the correct temperature, because it affects the reaction rate.
7
Q
What errors could be made?
A
- Measuring may be wrong.
- Starting and stopping timers may be faulty.
- A buffer solution should be used to keep the PH fixed.
8
Q
What are the hazards and risks?
A
- Don’t spill iodine on yourself.
- Be careful if a busen burner is used to heat the water bath.
