What is the lowry method?
The Lowry method is a combination of the biuret reaction and the reaction given by the Folin-Ciocalteu reagent (a complex of phosphomolybdic and phosphotungstic acids) with tyrosine, tryptophan and probably cysteine and histidine residues present in the protein. This results in blue-coloured products, whose absorbance is read at 750 nm. This allows the determination of proteins at concentrations of 1 ug/ml and above. Due to the differences in amino acid composition, different proteins give different colors with the Folin-Ciocalteu reagent, so it is important to use the corresponding standard for the protein under investigation.
