Chromosomes is start replicating and becoming visible as pairs of Chromatids. The chromosomes line up at the equator and the chromatids are pulled to the opposite Poles by spindle fibres to form new nuclei.

The cell cytoplasm splits between the new nuclei to form two daughter cells.

It requires aseptic techniques, in which growth of other cells such as bacteria is prevented and the culture remains uncontaminated.

It requires an appropriate liquid or solid medium such as nutrient agar or Bros in which the cells can grow and divide.

Temperature, pH and oxygen levels.
