polarizing microscopy Flashcards
what is polarizing microscopy
allows light to bend in certain direction and gives off colors specific to certain crystals
-uses to diagnose crystals in urine and synovial fluid
-lipids in urine
can bend polarized light
anisotropic or birefringent
2 polarizing filters
polarizer and analyzer
-put both together and stops direction of light
polarizer
placed at bottom of microscope
-forces light to travel in left to right
analyzer
placed at head of microscope
-forces light to move north to south
birefringent material
-rotate direction of light that passes through
-appears bright against black background
if substance rotates plane of polarized light clockwise
positive birefringence
If substance rotates plane of polarized light counterclockwise
negative birefringence
to detect another component we add a
compensator
what does a compensator do
tells us which direction of light is bent in
-placed between analyzer and polarizer
has arrow to indicate direction
aligned 45 degrees
most common compensator
red
positive birefringent
-blue color when parallel to direction of compensator
-Yellow when perpendicular to compensator
negative birefringent
○ Yellow when parallel
-Blue with perpendicular
