ONPG test
Has 2 tubes filled with ONPG broth. Inoculate bacteria and add ONPG disk. Used to detect late lactose fermenters from the enzyme B-galactosidase in the cytoplasm.
Urea Hydrolysis/ Urease test
2 tubes filled with 2mL water, inoculated with bacteria and urease disk. Used to differentiate organisms that have ability to hydrolyze urea with enzyme urease.
Phenylalanine Deaminase test
2 tubes filled with agar in a slant. Inoculated by dragging bacteria. Used to identify bacteria with the enzyme phenylalanine deaminase which removes an amine group
Starch Hydrolysis test
1 starch plate divided in half. Used to differentiate bacteria with ability to hydrolyze starch with enzyme Amylase.
MR test
2 tubes inoculated with bacteria. Used to identify bacteria that produce stable acid end products by mixed-acid fermentation of glucose.
VP test
2 tubes inoculated with bacteria. Used to identify organisms able to produce acetone from degradation of glucose.
Decarboxylation test
2 tubes with amino acid broth and layer of mineral oil. Used to detect if organism can remove the carboxyl group from amino acid.
Simmon’s Citrate test
2 tubes (citrate slants). Used to determine the ability of an organism to use citrate as its sole carbon source.
Tryptophan Hydrolysis/ Indole Production test
2 tubes of tryptone broth. Used to identify bacteria capable of producing indole using the enzyme tryptophanase.
Kliger’s Iron Agar (KIA) test
4 slants requiring stab and streak. Differentiates bacteria on the ability to ferment glucose and lactose, produce gas or no gas, or produce hydrogen sulfide.
O-F/ Oxydation-Fermentation test
4 tubes of O-F glucose. Used to differentiate bacteria on ability to oxidize or ferment specific sugars. 2 of the tubes are sealed with mineral oil to provide anaerobic conditions.
Nitrate Reduction Test
3 tubes of nitrate broth. Used to detect the ability of an organism to reduce nitrate NO3–>NO2 or some other nitrogenous compound using the enzyme nitrate reductase. Includes following 3 steps for conformation.
Sugar Fermentation test
3 tubes with sugar and reagent phenol red which allows determination of a species fermentation end products and ability to enzymatically convert other sugars into glucose. Has Durham tube: Bubbles= gas production
