Pharmacokinetics is ..
what the body does to a drug (drug movement into and throughout the body)
Invasion
active substance is freed from dosage form, absorption from administration site, distribution into human body
Elimination
Metabolism (biotransformation), excretion of drugs
Absorption depends on
the drug’s physicochemical properties, the formulation, the route of administration.
Passive diffusion
high to low concentration (cannot go against it), no energy, rate depends on gradient, solubility (lipids more rapid), size (smaller more rapid), ionization (unpolarized more easily), area of absorption (small intestine large area)
Facilitated diffusion
drug combines with some carrier molecules, no energy, can go against, only for similar to endogenous substances.
Pinocytosis
membrane invaginate, encloses fluid or particles, form vesicle in interior, requires energy
Active transport
needs carriers, does require energy, transport against gradient, specific only for drugs structurally similar to endogenous.
