MOA of the non benzo hypnotic?
Selective receptor binding to BNZ receptor 1, which is responsible for sedation
What are three non-benzo hypnotics usually used for sleep?
- Zolpidem
- Zaleplon
- Eszopiclone
What is the indication to use zolpidem, zaleplon or eszoplicone?
Short term use as sleep aid. Not as much issue with tolerance/dependance, but can still happen.
Order of half-life from shortest to longest
Zaleplon < Zolpidem < eszopiclone
4 side effects of the non benzo drugs?
- anterograde amnesia
- hallucinations
- sleepwalking
- GI side effects (most common)
What is the MOA of Ramelteon? Benefit of using?
selective melatonin MT1 and MT2 agonist
*no tolerance or dependence
What is MOA of buspirone?
partial agonist of the 5HT-1A receptor
*does not potentiate the CNS effects of alcohol so is safe for alcoholics; also not as high risk for abuse as benzos, but takes 1-2 weeks to work
What drug is buspirone given with commonly for anxiety and why?
Not as effective, so often used with SSRI
Other than buspirone, what would be a good anti-anxiolytic to give someone that cant take benzos?
Hydroxyzine - is an anthistamine and has anticholinergic effects
