L-Dopa
Most effective for parkinson; Dopamine pre-cursor; coadminister with Carbidopa
Adverse: n/v, hallucinations, compulsive behavior, dyskinesia
End of dose deterioration & on-off phenomenon
Proamipexole & Ropinirole
D2 Agonist
Late stage of PD; same side effects as L-Dopa
Benzotropine & Diphenhydramine
Anticholinergics (muscarine agonist)
better for tremor & rigidity than bradykinesia
adverse: drowsy, mood change, blurred vision, memory loss
Less effective then L-Dopa and D agonist; for neuroleptic induces Parkinsons
Selegiline & Rosagiline
MAO Inhibitors (irreversible inhibitors)
MAO-B responsible for DA metabolism
much safer than MAO-AI (anti-depress)
take with L-Dopa later, neuro-protective
Entacapone
COMT inhibitors
restricted to periphery to elevate L-Dopa
Amantadine
Anitiviral drug; use limited to 6-8 months
NMDAR inhibition; Increase DA; anticholinergic
early, mild stage of PD
Baclofen
Spasmolytic
GABA-B receptor agonist ; MS, ALA, SC Injury
Benzodiazepines
GABA-A receptor agonist; relief acute muscle spasm
Dantrolene
inhibit ryanodine recepter & diminish Ca release from sarcoplasmic reticulum
IV: malignant hyperthermia & neuroleptic syn.
Oral: MS & cerebral palsy
