MOUNTING Flashcards
A syrupy fluid applied between the section and the coverslip after staining, setting the section firmly, preventing the movement of the coverslip.
Mounting Medium
To protect stained section from physical injury and from bleaching or deterioration of the stain as a result of oxidation.
Mounting Medium / Mountant
For best result stained tissue sections must be impregnated by a transparent medium with a refractive index of
1.518
TRUE OR FALSE
Mounting medium should be colorless and transparent.
TRUE
TRUE OR FALSE
It should have a refractive index
close to that of diamond
FALSE
It should have a refractive index
close to that of GLASS
TRUE OR FALSE
It should be freely miscible with xylene and toluene.
TRUE
TRUE OR FALSE
It should not dry to a non-stick consistency and soften relatively quickly
FALSE
It should not dry to a non-stick consistency and harden relatively quickly
TRUE OR FALSE
It should not cause shrinkage and distortion of tissues
TRUE
Designed to mount water-miscible preparations directly from water in cases where the stain is removed or decolorized with alcohol or xyelene
Aqueous / Water soluble mountants
Low refractive index
Water
good only for temporary mounting
Water
also used as a preservative
Glycerin
has high index of refraction
Glycerin
Refractive Index of Glycerin
1.47
does not solidify upon storage
Farrant’s medium
RI of Gum syrup
1.43
(i love Gum)
used for methylene blue stained nerve preparations and as general purpose aqueous mountant
Apathy’s medium
recommended for mounting frozen sections from water or paraffin sections which require dehydrating and clearing
Brun’s fluid
used for preparations that have beeen dehydrated and cleared in xyelene or toluene and are commended for majority of staining methods
Resinous mountants
natural resin extracted from the Canadian Tree, Abus balsamea
Canada Balsam
recommended for whole mounts and for thick sections because it does not shrink much
Canada Balsam
Miscible with xyelene and is quite expensive
Canada Balsam
RI of Canada Balsam
1.524
recommended for small tissue sections
Dibutyphthalate polystrene xylene (DPX)
RI of DPX
1.532
RI of XAM
1.52
synthetic resin mixture in xylene in pale yellow or colorless solution
XAM
RI of Clarite
1.544
synthetic resin hich is soluble in xylene
Clarite
PERMANENT MOUNTING MEDIA
- Canada Balsam
- D.P.X
- Clarite
- Eukitt
- Entellan
this dissolves out the essential staining
Xylol
For some preparations, it is imperative not to use the permanent _ mounting media
Xylol-resin
this dissolves out the essential staining
SEMI-PERMANENT AND TEMPORARY MOUNTING MEDIA
- Water
- Glycerin
- Mineral Oil
- Glycerin jelly
- Von Apathy’s Gum syrup medium
- Farrant’s medium
- Levulose (Fructose) Syrup
