Devise a method to investigate the effect of pH on amylase enzyme activity.

A
  • Heat water to a constant 35°C
  • Put two drops of iodine solution into each well of spotting tile
  • Add 2cm³ of amylase enzyme solution to a test tube
  • Place 2cm³ of starch solution into same tube
  • Add 1cm³ of buffer solution with a pH of 5 to the tube
  • Mix solution in test tube and place it into beaker of water
  • Use pipette to remove few drops of solution every 20 seconds from test tube and put them into a different well of spotting tile
  • Repeat until iodine solution stops turning black
  • Record the time this takes
  • Repeat with buffer solutions of different pH values to see how pH affects time taken for starch to be broken down
