Module 1.3 - Phytochemical Tests (Cardenolides and Bufadienolides) Flashcards
Positive result for Keller-Killiani test
Purple color at the junction
Remarks for Keller-Killiani test
Presence of 2 - deoxysugar
Positive result for Lieberman-Burchard test
Colors ranging from blue to Green, red, pink, purple or violet
Remarks for Lieberman-Burchard test
Presence of steroid/triterpenoid skeleton
Positive result for Salkowski’s test
Red color
Remarks for Salkowski’s test
Presence of unsaturated sterol and/or triterpenes
Process for Keller Killiani test
- Extract with chloroform, evaporate to dryness
- Add 0.4 mL of glacial acetic acid containing a trace amount of ferric chloride
- Transfer to small test tube, ass 0.5 mL of conc. Sulphuric acid by the side of the test tube
- Blue color appears in the acetic acid layer
Process for Libermann-Burchard test
- 2 mg of dry extract was dissolved in Acetic anhydride
- Heated to boiling
- Cooled
- add 1 mL of conc. Sulphuric acid was added along the sides of the test tube
- Formation of a pink/red color indicates triterpenoids
Positive result for Liebermann-Burchard’s test for STEROIDS
Green color
Process for Liebermann-Burchard test for STEROIDS
- 2 mg of dry extract dissolved in Acetic anhydride
- Heated to boiling
- Cooled
- 1 mL of conc. Sulphuric acid added to sides of test tube
Positive result for Salkowski’s test for STEROIDS
Red color
Process for Salkowski’s test for STEROIDS
- 2 mg of dry extract was shaken with chloroform
- Sulphuric acid slowly added to the chloroform layer by the sides of the test tube
