Microscopy Flashcards
Whats microscopy used to do?
Study very small objects
To look at cells or organelles
What do light microscopes use
An external light source
Light microscope negatives
Lower magnification
Lower resolution
Difficulty distinguishing
Does not zoom in as much
Light microscope advantages
Cheaper
Can see whole cells nuclei and vacuoles
What do electron microscopes use?
Electrons to generate the images
Electron microscope advantages
Magnification much higher
Resolution much higher
Used for much smaller things like ribosomes
Disadvantage of electron microscopes
Expensive
How to prepare a light microscope
1) take a slide and put a small water droplet on it
2) take a thin layer of your specimen so the light goes through
3) use a stain (a chemical that you put on the specimen) this highlights specific organelles
4) place a cover slip on top and avoid bubbles
How to calculate magnification
Image size= magnification x actual size
