What is confocal microscopy used for?
Optical sections via a laser to show 3D structure - illuminates and eliminates out ‘focus flare’
When is dark field microscopy used?
Image is scattered light rather than transmission - used to detect syphilis and malaria
What is phase contrast microscopy used for?
Viewing detail in unstained living cells - heightened contrast
What is fluorescence microscopy used for?
Can see where individual substances/structures lie in a cell
What does haematoxylin stain?
Acidic components - purple blue e.g. DNA
What does eosin stain?
Basic components - pink e.g. cytoplasmic proteins
What does periodic acid shiff stain?
Magenta stain - Carbohydrates and glycoproteins
Briefly describe the fixation process
Water, ethanol, xylene, embedding in wax, sectioning, xylene, ethanol, water, staining, dehydration.
